The COBAS AmpliPrep/COBAS TaqMan HIV-1 is an in vitro nucleic acid amplification test for the quantitation of Human Immunodeficiency Virus (HIV-1) nucleic acid in human plasma that targets two highly conserved regions of the HIV-1 genome, not subject to drug pressure, using the COBAS AmpliPrep Instrument for automated sample preparation and the COBAS TaqMan Analyzer or COBAS TaqMan 48 Analyzer for automated amplification and detection. This test is intended for use in conjunction with clinical presentation and other laboratory markers of disease progress for the clinical management of HIV-1 infected patients.
   

 

 

 

Working Principle

  The COBAS AmpliPrep/COBAS TaqMan 48 HIV-1 Test is based on three major processes:
1. Automated specimen preparation to isolate HIV-1 RNA — COBAS AmpliPrep Instrument
2. Automated reverse transcription of RNA and Polymerase Chain Reaction (PCR) amplification of the resultant cDNA — COBAS TaqMan 48 Analyzer
3. Automated detection of amplified cDNA (amplicon) using cleaved dual fluorescent dye-labeled oligonucleotide detection probes.
By including an HIV-1 Quantiation Standard (QS) at a known copy number, it is possible quantitate HIV-1 in real time —COBAS TaqMan 48 Analyzer

     
User Instructions   4 ml of whole blood collected in K3 EDTA should be transported to the Department of HIV/AIDS, NIRT within 4 hours from the time of collection at room temperature
